TNFa is presumably shed from cell membranes by TNFa-FW, cleaving enzyme (TACE). Incubation of the TACE substrate with recombinant human TACE gives a specific cleavage to restore the quenched fluorescence. The substrate is widely used to screen inhibitors of TNF-α converting enzyme (TACE, ADAM17 endopeptidase) activity.
