肝臟星形細(xì)胞是小葉內(nèi)結(jié)締組織細(xì)胞,呈現(xiàn)肌成纖維細(xì)胞樣或脂肪細(xì)胞形態(tài)。他們參與肝臟細(xì)胞外基質(zhì)的穩(wěn)態(tài),修復(fù),再生,纖維化以有控制視黃醇的代謝,存儲(chǔ)和釋放。肝臟受傷后,肝臟星形細(xì)胞轉(zhuǎn)化成肌成纖維細(xì)胞樣,并且是纖維化的肝臟中 I 型膠原的主要來(lái)源。除了以上特征,已發(fā)現(xiàn)在疾病狀態(tài)和肝內(nèi)門(mén)靜脈高壓的發(fā)病過(guò)程中,肝星狀細(xì)胞通過(guò)細(xì)胞收縮調(diào)節(jié)肝臟微循環(huán)[1,2]。星形細(xì)胞的增殖、遷移和趨化因子的表達(dá)參與了肝臟炎癥和纖維化的發(fā)病過(guò)程。因此,星形細(xì)胞激活的分子調(diào)控在將來(lái)有助于治療肝纖維化,并且能減少慢性肝病患者的發(fā)病率和死亡率。
Sciencell研究實(shí)驗(yàn)室的人肝星形細(xì)胞是從人肝臟組織中分離得到的,原代凍存,冰凍運(yùn)輸。每管細(xì)胞密度*過(guò)5×10^5/ml。人臍靜脈平滑肌細(xì)胞經(jīng)desmin 和α-actin免疫熒光鑒定。本細(xì)胞經(jīng)檢測(cè)不含HIV-1、HBV、HCV、支原體、細(xì)菌、酵母菌和真菌。如采用ScienCell 實(shí)驗(yàn)室特制的培養(yǎng)基,可保證此細(xì)胞15倍增增殖。? ?
