少突膠質(zhì)前體細(xì)胞是Raff、Miller 和Noble于1993年**發(fā)現(xiàn)[1],并已進(jìn)行了深入的研究。在文獻(xiàn)中,前體細(xì)胞指少突細(xì)胞型星形膠質(zhì)細(xì)胞祖細(xì)胞或少突膠質(zhì)細(xì)胞前體細(xì)胞。發(fā)育中和成熟的**神經(jīng)系統(tǒng)都含有少突膠質(zhì)前體細(xì)胞[2, 3]。少突膠質(zhì)細(xì)胞,**神經(jīng)系統(tǒng)中的髓磷脂組成細(xì)胞,來源于少突前體細(xì)胞。在培養(yǎng)中,少突前體細(xì)胞在基本的成纖維細(xì)胞生長因子存在的情況下可產(chǎn)生于神經(jīng)祖細(xì)胞或神經(jīng)干細(xì)胞,OPC在血小板衍生的生長因子或星形膠質(zhì)細(xì)胞產(chǎn)生的生長因子[4]存在的情況下才能增殖,并分化成為成熟的少突膠質(zhì)細(xì)胞。正因如此,它們?yōu)檠芯堪l(fā)育轉(zhuǎn)型提供了特殊的細(xì)胞種群。
ScienCell實驗室的HOPC分離自人腦,原代凍存,冰凍運輸。每管細(xì)胞密度*過1×106/ml。細(xì)胞經(jīng) A2B5和O4免疫熒光鑒定。本細(xì)胞經(jīng)檢測不含HIV-1、HBV、HCV、支原體、細(xì)菌、酵母菌和真菌。如采用ScienCell 實驗室特制的培養(yǎng)基,可保證此細(xì)胞生長良好,此細(xì)胞不推薦增殖培養(yǎng)。
