ELISA方法的基本原理是酶分子與抗體或抗抗體分子共價(jià)結(jié)合，此種結(jié)合不會(huì)改變抗體的免疫學(xué)特性，也不影響酶的生物學(xué)活性。此種酶標(biāo)記抗體可與吸附在固相載體上的抗原或抗體發(fā)生特異性結(jié)合。滴加底物溶液后，底物可在酶作用下使其所含的供氫體由無(wú)色的還原型變成有色的氧化型，出現(xiàn)顏色反應(yīng)。因此，可通過(guò)底物的顏色反應(yīng)來(lái)判定有無(wú)相應(yīng)的免疫反應(yīng)，顏色反應(yīng)的深淺與標(biāo)本中相應(yīng)抗體或抗原的量呈正比。此種顯色反應(yīng)可通過(guò)ELISA檢測(cè)儀進(jìn)行定量測(cè)定，這樣就將酶化學(xué)反應(yīng)的敏感性和抗原抗體反應(yīng)的特異性結(jié)合起來(lái)，使ELISA方法成為一種既特異又敏感的檢測(cè)方法。
